Punicalagin is a large-molecule ellagitannin polyphenol derived from pomegranate, and it is the most abundant polyphenol active component in pomegranate. Its molecular backbone consists of glucose linked to multiple hexahydroxydiphenoyl (HHDP) structural units. After ingestion, it undergoes hydrolysis mediated by gastric acid, digestive enzymes and gut microbiota. The HHDP units are converted into ellagic acid via lactonization; each molecule of punicalagin can yield 2-3 molecules of ellagic acid. Due to its large molecular size, punicalagin can hardly be directly absorbed into the bloodstream.

As a key intermediate metabolite from in-vivo hydrolysis, ellagic acid is further transformed into urolithins by gut microbiota. These urolithins are the metabolites that enter systemic circulation and exert physiological effects. In short, punicalagin and ellagic acid have a precursor-hydrolysis-product relationship. Punicalagin shows better water solubility than ellagic acid, while ellagic acid has a smaller molecular weight and serves as a vital intermediate form for biological activity in the human body.

In food supplements, extracts containing 30-40% punicalagin are not classified as novel foods and may be used without pre-market authorisation. However, when used in conventional foods other than food supplements (e.g. beverages and snacks), such extracts are regarded as novel foods and require prior authorisation from the European Commission before being legally placed on the market, as pomegranate pericarp has not been significantly consumed as a traditional food in the EU. Furthermore, in the field of feed additives, EFSA has received an application for authorisation of pomegranate peel extract (40% punicalagin) as a sensory additive for all animal species, which is currently under assessment.

Pomegranate peel extract is classified as a dietary supplement raw material and does not require pre-market approval by the FDA. Punicalagin does not hold an independent GRAS status. High-purity monomeric punicalagin has no publicly available NDI notification and cannot be directly marketed as a dietary ingredient; manufacturers are required to submit an NDI notification. In cosmetics, the CIR has approved the use of pomegranate peel extract with no usage restrictions.
