Mangiferin Solubility

 

Mangiferin is a polyphenolic glycoside with moderate overall polarity. Its solubility varies significantly with solvent polarity and temperature. At room temperature, it is only slightly soluble in moderately polar organic solvents, and its solubility increases markedly upon heating. It exhibits the best solubility in DMSO, a strongly polar aprotic solvent, and a clear concentrated solution can be obtained with the assistance of ultrasonication. It is sparingly soluble in methanol and ethanol, while its dissolution is greatly improved in hot dilute methanol and hot dilute ethanol. It is practically insoluble in weakly polar and nonpolar organic solvents such as diethyl ether, chloroform, petroleum ether and n-hexane, and barely soluble in acetone and ethyl acetate. Overall, its solubility follows the rule that higher solvent polarity and higher temperature lead to better dissolution performance.

Mangiferin has extremely poor water solubility at room temperature. Its saturated solubility in pure water at 25 °C is less than 0.1 mg/mL, making it practically insoluble in cold water. High-concentration aqueous mangiferin solutions cannot be formulated with pure water alone.

USA

In the United States, low-purity mangiferin extracts from mango leaves can be freely circulated as botanical dietary supplement raw materials, and enterprises may complete self-affirmed GRAS filings for use in functional foods, sports nutrition capsules and tablets. Although there is no official GRAS notice for high-purity mangiferin monomer, it also qualifies for the self-affirmed GRAS pathway. There is no legally mandated maximum daily intake limit, while the generally accepted safe industry dosage of pure mangiferin does not exceed 500 mg per day. For cosmetic applications, it is a compliant botanical active ingredient with antioxidant and soothing effects. From a chemical regulatory perspective, mangiferin is not listed on the TSCA Inventory.
