Starting Materials and Reagent Preparation

Indole was used as the starting material. N,N-dimethylformamide (DMF), phosphorus oxychloride (POCl₃), sodium borohydride (NaBH₄), ethanol, dichloromethane, sodium hydroxide, dilute hydrochloric acid, and pure water were prepared. The reaction equipment and vacuum drying oven were ensured to be clean and in working order.

 

Formylation Synthesis of Indole-3-Carbaldehyde

DMF was added to a dry reaction vessel and stirred while cooling. Phosphorus oxychloride was then slowly added dropwise, followed by continued stirring to form the Vilsmeier reagent. Indole was then dissolved in a small amount of DMF and slowly added dropwise to the reaction solution. After cooling to room temperature, the reaction solution was slowly poured into an ice-water mixture for hydrolysis. The pH was adjusted to 7-8 with 30% sodium hydroxide solution, resulting in the precipitation of a large amount of yellow solid. This solid was filtered, washed with water until neutral, and dried to obtain crude indole-3-carbaldehyde.

 

Synthesis of Crude 3-Indolemethylethanol

Indole-3-carboxaldehyde was dissolved in methanol or ethanol and stirred until completely dissolved. The solution was placed in an ice bath at 0–5°C, and sodium borohydride was added in batches. After the reaction was complete, dilute hydrochloric acid or saturated ammonium chloride solution was slowly added to quench excess reducing agent and adjust the pH to 7. Most of the alcohol solvent was then removed by vacuum distillation. The residue was extracted 2–3 times with ethyl acetate or dichloromethane. The combined organic phases were dried over anhydrous sodium sulfate, filtered, and then distilled under vacuum to obtain a white to off-white crude 3-indolemethylethanol.

 

Purification to Obtain Final Product

The crude 3-indolemethylethanol was dissolved in a suitable amount of hot ethanol-water mixed solvent until just dissolved. The solution was filtered while hot to remove insoluble matter. The filtrate was slowly cooled to room temperature and then placed in an ice bath to allow crystals to settle. The crystals were collected by vacuum filtration and washed with a small amount of cold solvent. Finally, the solution was dried under vacuum at 40–50°C to obtain a high-purity 3-indolemethylethanol product with a purity of over 99%.
