How to make Deazaflavin Powder?

Condensation Reaction
Charge barbituric acid and 2-nitrobenzaldehyde into the reactor at a 1:1.2 molar ratio, add water and isopropanol as mixed solvent, heat to 82°C, and reflux for 2-6 hours to generate the yellow benzylidene barbituric acid intermediate. After the reaction is complete, cool down, filter under suction, and dry to obtain a yellow solid powder with a yield of approximately 86%.
Reduction and Cyclization
Add the intermediate from Step 1 to a mixed solvent of water and tetrahydrofuran, heat to 70°C, and add a sulfide reducing agent in batches to reduce the nitro group to an amino group, which immediately undergoes nucleophilic attack to complete cyclization and form the core deazaflavin skeleton. After approximately 5 hours of reaction, concentrate, filter under suction, and wash with water to obtain the crude product.
Purification
Add the crude product to isopropanol as solvent, heat to 88°C, and stir for 3 hours to slurry. Filter hot to remove insoluble impurities, then cool to crystallize. Separate the crystals by suction filtration and dry to obtain a white pure powder with purity exceeding 99% .
